Uji In Silico Interaksi Senyawa Tropis Alami dan Sintetik Terhadap Protein Bcl-2 Sebagai Agen Antikanker Payudara
In Silico Study of Tropical and Synthetic Compounds Targeting Bcl-2 Protein as Breast Cancer Therapeutic Agents

Penyebab kematian utama bagi perempuan secara global adalah penyakit kanker payudara (breast cancer). Resistensi terhadap kemoterapi dan efek samping pengobatan konvensional mendorong pencarian agen terapi alternatif berbasis senyawa alami. Salah satu target molekuler yang penting dalam mekanisme apoptosis sel kanker adalah protein Bcl-2 dan keluarganya. Studi ini bertujuan meninjau secara sistematis hasil molecular docking in silico senyawa alami dari tanaman tropis dan sintetik terhadap protein Bcl-2 sebagai target pengembangan terapi kanker payudara. Sebanyak 21 artikel orisinal lima tahun terakhir dianalisis berdasarkan sumber senyawa (tanaman atau sintetik), senyawa aktif, kode protein (PDB), serta nilai afinitas ikatan. Hasil menunjukkan bahwa sebagian besar senyawa memiliki potensi sebagai inhibitor Bcl-2 dengan nilai afinitas yang kuat, antara lain Niaziminin B dari Moringa oleifera (−14.96 kcal/mol) sebagai salah satu yang tertinggi. Tinjauan ini menegaskan potensi senyawa alami dari tanaman tropis dan sintetik sebagai kandidat terapi kanker payudara melalui mekanisme inhibisi Bcl-2. Uji lanjutan seperti uji in vitro dan uji in vivo diperlukan untuk memastikan aktivitas biologis serta toksisitas dari berbagai senyawa alam maupun sintetik dalam pengembangan obat.
